Small molecule-mediated allosteric activation of the base excision repair enzyme 8-oxoguanine DNA glycosylase and its impact on mitochondrial function

The authors of this study conducted a mechanistic assessment of enhanced 8-oxoguanine DNA glycosylase (OGG1) catalytic activity coupled with molecular modelling of potential ligand binding sites, as well as biophysical measures of OGG1 activators. The CytoSMART Exact (formerly known as the Corning Cell Counter) was used to count mouse embryonic fibroblasts (MEFs) before plating them and performing further assays. The results of the study support an allosteric mechanism of OGG1 and not product-assisted catalysis.
